超高24平方深孔板孔内流体力学
当培养体积为2.5~3.5 mL(大可用于常规24平方深孔板)时,应用超高24平方深孔板(井体积:25毫升)是不够的。是在转速为300转,振幅为50毫米的振动筛上,氧转移率在40至60 mmol O2 l-1 h-1之间(对应于200-300 h-1的kLa值)是容易达到的
建议(默认)条件1:25毫米振动振幅,375转/分(NB上大肠杆菌培养,20倍速度减慢)5毫升6毫升7毫升8毫升
Hydrodynamics inside wells of extra-high 24 square deepwell plates
Extra-high 24 square deepwell plates (well volume: 25 ml) are applied when culture volumes of 2.5 to 3.5 ml (as can be maximally used for regular 24 square deepwell plates) do not suffice.With extra-high 24 square deepwell plates, culture volumes up till 8 ml can be used, preferably at 300 rpm on a shaker with a 50 mm amplitudeOxygen-transfer rates between 40 and 60 mmol O2 l-1 h-1 (corresponding to kLa values of 200-300 h-1) are readily attainable.For the (slowed-down) high-speed videos below we used hitplates from HJ- Bioanalytik
Adviced (default) condtion 1:25 mm shaker amplitude, 375 rpm(E. coli culture on NB, 20 times slowed down) 5 ml 6 ml 7 ml 8ml
