Glycotech31-010,Glycotech31-001平行板流动小室(Parallel-Plate Flow Chamber),Rectangular Flow Chamber Kit,Circular Flow Chamber Kit

平行板流动小室Parallel-Plate Flow Chamber)是体外研究细胞力学zui常用的模型之一,用来模拟体内各种细胞处于生理环境中,暴露在动态液体流作用下所受到的流体剪应力。体外细胞的代谢反应与所受到的壁面剪应力密切相关。

典型的平行板流动小室由三个部分组成:聚碳酸酯分液室,硅胶垫圈和玻璃盖玻片。分液室构成流动小室的一侧,包括进液口,出液口和真空槽。垫圈的厚度决定流程长度。盖玻片构成流动小室的另一侧,其上覆盖细胞外基质蛋白(ECM),血管细胞或其他待研究的生物材料。真空形成一个密封环境将三个部件紧密连接在一起,同时也确保通道高度的一致性。

优 势:

1.平行板流动小室能够产生生理学范畴内的壁面剪应力:0.01-30 dyn/cm2。通过注射泵以可控的运动力将液体(如抗凝的全血或者细胞悬液)注入小室并流进固定化基质的方法产生剪应力。

2.装置设计,组装和操作简单,方便;

3.可以在设定的时间-周期内研究稳定剪应力对细胞的影响;也可以随实验需求调节流速和剪应力;

细胞可在流动条件下生长,能够在显微镜下随时观察生长状态;或者利用视频显微镜实时监控细胞状态;

应 用:

1.模拟体内血液流动产生的血流剪切力,研究不同剪应力下白细胞和内皮细胞之间的动态粘附作用或白细胞受体-配体间的相互作用等;

2.利用平行板流动小室产生剪应力模拟肿瘤细胞的体内生长环境,从而研究肿瘤细胞的增殖,吸附和转yi机制。

3.细胞趋化实验中作为药物测试体系,基于白细胞-内皮细胞粘附过程作为新药物的运输体系;

Static Cell Adhesion Wash Chamber, 货号:31-006The use of convenient multiwell plates to assay the adhesion of cells to immobilized molecules under static conditions is usually difficult due to the uncontrolled shear forces normally generated while washing the plate.

The Wash Chamber allows the wells of the multiwell plate to be washed by inversion in a liquid-filled environment. Unbound cells are separated from the wells by gravity thereby eliminating shear and the passage of the adherent cells through a liquid/air interface.(Refer to Protocol 8)

CELL ADHESION CHAMBER for TLC Plates, 货号:31-007Cell surface carbohydrates act as specific recognition molecules for intercellular adhesion among certain cells.

Glycosphingolipids represent a group of cell surface glycoconjugates that are easily separated and analyzed by thin layer chromatography. The cell adhesion chamber was specifically designed to permit the assay of cell adhesion directly on glycosphingolipids separated on thin layer chromatograms.

Cells are centrifuged directly on the TLC plate to form immediate close contact with chromatographed glycosphingolipids. The position of the bound cells corresponds to the Rf value of the glycosphingolipid ligand which may be used for purification and structural characterization of the functional carbohydrate sequence.(Refer to Protocol 9)
