使用抗原设计软件选择与其他人类蛋白同源性低50-150个氨基酸Protein Epitope Signature Tag (PrEST)序列。避免了使用全序列重组蛋白造成的与其他蛋白有高同源区域造成的非特异性。
※ Protein Epitope Signature Tags (PrESTs)
● PrESTs是蛋白质组研究项目Human Protein Atlas Project（HPA) 开发的抗原标签，这种标签来自目标蛋白的的遗传序列，其准确性可由质谱进行验证。
● PrESTs的选择 选择PrESTs的准则是“目标蛋白中与其它蛋白序列相似性小的片段”。为了达到这一个标准，采用了严格的生物信息学判别程序，利用BLASTP对ENSEMBl数据库记录的所有蛋白序列进行了扫描、打分和筛选。
● PrESTs生产与验证 PrESTs通过大肠杆菌大量表达，产物将通过严格的结构解析，以确认其准确性。
